Six Senses Douro Valley occupies a restored 19th-century manor at Quinta de Vale Abraão, on a hillside above Samodães, close to Lamego. The building sits over vine-covered terraces that run down to the Douro river below. The original structure has been refurbished with contemporary interiors that keep traces of the region's architecture, and the estate includes flourishing kitchen gardens and working vineyards.

There are 71 guest accommodations across the property, from Quinta and Vineyard rooms and suites in the main building to Valley Rooms and Suites housed in a separate renovated wine lodge next door. The nine Valley units can be booked individually or combined for groups of up to nine bedrooms, sharing a lounge with a fireplace, gazebo-shaded gardens, and a heated pool with vitality pool. For larger parties there is also a One or Two Bedroom Villa tucked into the gardens, each with its own private swimming pool. Dining draws on produce grown in two organic gardens, one on the estate and one at nearby Quinta do Mourão, served across the Dining Room and Open Kitchen, Cozinha do Douro, the Quinta Bar and Lounge, and a seasonal Summer Garden that runs from June to September. The Wine Library holds tastings guided by the resort's sommelier, and destination dining sets picnics and meals by the river, in the vineyards, or in a Georgian-era grotto.

Six Senses Spa Douro Valley has ten treatment rooms, an indoor pool with underwater sound and colour therapy, a separate vitality pool, a vitality suite with several types of sauna and steam room, a fitness centre, studio, cryotherapy and an alchemy bar. Guided wellness programs cover sleep, yoga, stress relief and detox. Outside the hotel, activities include river cruises, grape stomping in season, mountain biking, tree climbing, water sports and visits to prehistoric rock art sites.
